RhoA Is a Negative Regulator of AR Function Because Akt can transactivate the AR in LNCaP cells, we were interested to determine whether inhibition of Akt activity by RhoA can modulate AR function. LNCaP cells were transfected with AR reporter plasmid along with the constitutively active form RhoA G14V ; or the dominant-negative form RhoA T19N ; of RhoA GTPase Fig. 11B ; . Constitutively active RhoA was able to reduce the transactivation functions of AR, whereas the dominant-negative form of RhoA enhanced the androgen-dependent transactivation functions of AR Fig. 11B ; . These results suggest that RhoA can also negatively regulate prostate cell growth by exerting its effect on AR signaling.

A limitation of the standard design of phase I trials is the difficulty in determining antitumor activity, especially with single agents. The evaluation of antitumor activity is not the primary objective of a phase I trial; however, antitumor response observed in a phase I trial, along with preclinical data, is used to direct future phase II studies.1 This is inherently difficult in a standard phase I trial, in which patients with a wide range of tumor types are enrolled and, in most cases, heavily pretreated. In addition, the entry criteria for phase I studies, such as those that require only two or three prior treatment regimens, may preclude enrollment of patients with certain tumor types, such as breast cancer.8 The phase I II trial may overcome the limitations of the standard phase I trial. In the phase I II study design, the MTD is determined in a group of patients with the same disease and may provide a more accurate measure of antitumor activity in that situation. The selection of tumor type is based on preclinical data and initial phase I studies.8, 9 In addition, more patients are enrolled at the MTD to further assess activity before moving into larger phase II III trials. This approach has been used in the early development of several chemotherapeutic agents. One possible limitation is the extra time required to accrue patients with the same tumor type.8, 9.

Seen after medium-depth chemical peels or dermabrasion, has also been described.50, 51 The ultrastructural changes seen in our small group of patients at 30 and 90 days after laser resurfacing appear to correlate with these histologic findings. It is known that ablation of photodamaged epidermis and upper dermis, whether by chemical or physical means, allows reepithelialization from deeper, less photodamaged cells, resulting in the restored structural and functional integrity of epidermal keratinocytes.26-38, 50 The increased number and organization of the tonofibrils in epidermal keratinocytes that we have seen at 30 and 90 days after laser treatment appears therefore to be significant, as these findings correlate with the normalization of keratinocyte differentiation from the basal layer to the stratum corneum seen histologically after laser resurfacing. Additionally, the loss of intracellular and intercellular epidermal vacuolization and the presence of folded and tightly apposed intercellular spaces of adjacent keratinocytes studded at intervals with innumerable desmosomes 30 and 90 days after treatment appear to be significant, as these findings are characteristically seen in normal squamous epithelium.54 Clinical studies have revealed a measurable and reproducible decrease in fine wrinkling and improvement in skin texture after laser resurfacing, but the mechanism by which these changes occur is not yet clear.39-48 Fitzpatrick et al39 have postulated that, when the carbon dioxide laser interacts with tissue, 3 zones of tissue damage are produced: a vaporized zone, a zone of irreversible thermal necrosis, and a zone of reversible thermal damage, in which collagen shrinkage is thought to take place. It has been documented that thermal damage to collagen itself results in shrinkage, but it is not clear how great a role this shrinkage plays in generating clinical improvement in wrinkles.55-57 It is also thought that repair of this layer during healing may account for tightening of sagging skin and improvement of creases.39, 46, 47 The clinically evident tightening of sagging skin after laser surfacing may be in part due to the formation of new collagen, the decrease in the amount of amorphous debris in the papillary dermis, and the increased activity and number of fibroblasts that have been documented histologically and ultrastructurally after laser and other resurfacing procedures.26-36, 38, 50 Nelson et al34 have in fact found that collagen's striation periodicity is reduced after medium-depth chemical peels, resulting in a more compact architecture, and that the diameter of individual fibrils is more variable, consistent with recent production of collagen by activated fibroblasts. Precise quantitative studies will be required to substantiate whether compression of the collagen bundles occurs immediately after laser resurfacing and whether the striation periodicity of the collagen is immediately changed after treatment. If, in fact, the striation periodicity of the collagen in the papillary dermis is reduced at 30 and 90 days after treatment, this may partially explain the clinical perception of tighter, smoother skin in patients treated with resurfacing lasers. The new deposition of dermal collagen that we have seen ultrastructurally after laser resurfacing seems to correspond to the papillary dermal repair zone de.

1. Understand sex differences in the mechanisms and processes involved in all phases of nicotine use and addiction. The group gave this recommendation the highest priority because of the many gaps in our understanding of sex differences in nicotine use and addiction. An examination is needed of the environmental, behavioral, genetic, molecular, cellular, neurobiological, and hormonal sources of variation and their interactions in animals and humans during all phases of nicotine addiction, including initiation, maintenance, withdrawal, and relapse. This recommendation encompasses a vast array of preclinical and clinical research. Preclinical. Animal models make it possible to examine the reinforcing effects of nicotine that are highly relevant to tobacco dependence in humans and that cannot easily be studied in human subjects--mainly for ethical reasons. In animals, potential behavioral effects of pharmacological agents can be more fully characterized. Furthermore, these animal models allow the investigation of the basic underlying behavioral and neurochemical mechanisms that are relevant to nicotine addiction for reviews, see 5, 18 ; . Behavioral studies of rodents suggest that females are more sensitive than males to repeated nicotine exposure, as demonstrated by a more rapid onset of nicotine-induced behaviors 23 ; . The reinforcing effects of nicotine have been examined in female rats. In one study, female rats self-administered nicotine and, when compared to males, showed a higher motivation to obtain nicotine because they acquired self-administration capabilities of low doses of nicotine, whereas male rodents did not 24 ; . In this study, different stages of the estrous cycle did not affect nicotine self-administration. However, since rodents have drastically shortened cycles 4-5 days in length ; , heterogeneity within each cycle stage i.e., animals reaching a stage of the cycle at different times ; may have limited the ability of the authors to find a relationship between the estrous cycle and the reinforcing effects of nicotine. Research should be conducted in animals that have pharmacologically synchronized estrous cycles to reduce variability in hormonal levels, thereby providing a more accurate picture of whether hormone levels can affect nicotine intake. This procedure has been used to establish a relationship between and mysoline.

The model's data and account structure closely follow the accounting conventions used in the input output studies of the U.S. economy by the Department of Commerce's Bureau of Economic Analysis. The comprehensive and detailed data coverage of the entire United States by county, along with the ability to incorporate user-supplied data at each stage of the model-building process, provides a high degree of flexibility in terms of both geographic coverage and model formulation. In applying the IMPLAN model to the plant, Exelon Corp. provided three basic types of data: purchase order expenditures by purchase order code, employee compensation expenditures and tax payment data for 2004. The purchase order data were mapped to IMPLAN's 528 sector codes in two ways. First, by identifying the largest contracts at each geographic level and assigning them an industrial classification code within IMPLAN sector codes. For the remaining expenditures, the data were mapped into IMPLAN codes based on average distributions obtained through detailed studies of six nuclear reactors. These purchase order data also were mapped into IMPLAN based on the areas where these purchases were made. Then the authors used an estimate of revenues from electricity sales into the wholesale market in 2004 to augment the purchase order and compensation data. This augmentation was necessary because purchase orders and compensation do not reflect all the economic value of the Exelon nuclear plants, while total output approximated by total revenues ; better reflects the full economic impacts of the plant. Revenue estimates were based on kilowatt-hours sold and the average wholesale price for electricity sold by Exelon's Pennsylvania nuclear plants during 2004. The estimated revenues were above the expenditure data provided by Exelon, indicating a nuclear generation profit margin that was incorporated into IMPLAN as profits associated with the operation of the plant. These data were then incorporated into the IMPLAN model, which combined specifics of the local economy with data on economic activity of the plants to provide estimates of the plant's total impacts. Once the data sets were complete, IMPLAN developed the economic impact estimates detailed in this report.

Income Taxes Deferred Federal and state income taxes are provided on all significant temporary differences between book basis and tax basis of assets and liabilities. Investment tax credits previously utilized for income tax purposes have been deferred on the Consolidated Balance Sheets and are recognized in book income over the life of the related property. Exelon and its subsidiaries file a consolidated Federal income tax return. Income taxes are allocated to each of Exelon's subsidiaries within the consolidated group based on the separate return method. Exelon estimates its income tax valuation allowance by assessing which deferred tax assets are more likely than not to be recovered in the future see Note 14--Income Taxes ; . Gains and Losses on Reacquired Debt Recoverable gains and losses on reacquired debt related to regulated operations are deferred and amortized to interest expense over the period consistent with rate recovery for ratemaking purposes. Gains and losses on other debt are recognized in Exelon's Consolidated Statements of Income as incurred see Note 6--Supplemental Financial Information.
Recognizing the value that the land provides both as a habitat and a resource, Exelon works to sustain its quality by addressing historical impacts, reducing present risks and impacts, and minimizing future threats. Regarding historical impacts, Exelon continues to remediate and close former manufactured gas plant mgP ; sites within its territories. During 2007, PECO completed this process at sites in Doylestown and Phoenixville. In Illinois, the Electric Power Research Institute recently recognized ComEd's mgP team for its participation in a joint study that helped revise Illinois soil cleanup standards. ComEd and PECO expect the majority of remediation at these sites to continue through at least 2015 and 2013, respectively. Exelon also continued its proactive evaluation of current and former company properties that may have been exposed to material releases during operations prior to the 1970s, when modern environmental laws were enacted. Over the course of 2007, ComEd and PECO together evaluated the risk at 100 substations and 26 service centers; no high-risk sites were identified. During 2007, Exelon made significant progress on its voluntary phase-out of equipment containing polychlorinated biphenyls PCBs ; . ComEd completed 106 equipment retrofills replacements and 930 PCB capacitor replacements, while PECO removed 490 PCB capacitors and retrofilled or removed 75 pieces of equipment. These were in addition to replacements completed by Exelon Nuclear and Exelon Power. Exelon remains on track to eliminate known PCB-contaminated equipment from its operations by year-end 2012. Exelon Nuclear cut its 2007 volume of low-level radioactive waste LLRW ; by 2 percent from 2006 through continued improvements in recycling and workforce education. These reductions were driven in part by the imminent loss of the Barnwell Disposal Facility in South Carolina as an option for storing Nuclear's LLRW. Exelon has on-site LLRW storage capacity at all of its nuclear plants. In 2007, Exelon's nuclear plants produced approximately 400 tons of used nuclear fuel. Exelon stores this fuel at our nuclear plants both in water-filled pools and in dry storage systems. Exelon continues to call upon lawmakers to develop and implement a cogent strategy for used nuclear fuel management. The company's interest in constructing new plants, such as the one we have proposed for Victoria County, Texas, remains contingent upon a satisfactory resolution of this issue. Meanwhile, progress on such efforts as the DOE's proposed national fuel repository in Yucca Mountain, Nev., remains slow. In the interim, Exelon will continue to store our used fuel at our sites something we can do safely through the extended operating lives of our plants. In December 2007, Exelon Nuclear announced that it will seek to accelerate the decommissioning of the company's Zion Station in Illinois. The company has contracted with Utah-based EnergySolutions to dismantle the nuclear plant, which stopped producing power in 1998, contingent upon approvals from the U.S. Nuclear Regulatory Commission NRC ; and the Internal Revenue Service. Used nuclear fuel currently stored at Zion would be moved to a dry cask storage facility at least 400 feet farther from the lakeshore than the station's current fuel pool. Such independent fuel storage facilities are licensed by the NRC and exist at 39 plants nationwide, including five of Exelon's nuclear stations. "This is a unique opportunity to make hundreds of acres of lakefront property available for other uses at least a decade earlier than we thought possible." Values for airway responsiveness and forced expiratory volume in 1 second did not differ significantly between the two groups. Adverse events immediately after treatment were more common in the bronchialthermoplasty group than in the control group but were similar during the period from 6 weeks to 12 months after treatment.

Alex Gorsky is the Company Group Chairman and Worldwide Franchise Chairman for ETHICON and a member of the Surgical Care Group Operating Committee, a role he assumed in February 2008. He obtained a Master's in Business Administration from The Wharton School of the University of Pennsylvania in 1996. Further delays in the nuclear waste program could prove costly under a settlement announced August 10, 2004, between the Department of Justice and Exelon Corporation, which had filed a breach-of-contract suit over DOE's failure to begin accepting spent fuel by 1998 as required by NWPA. Under the settlement, Exelon is to be reimbursed from the federal Judgment Fund for its spent fuel storage costs caused by the waste program delays. Exelon estimates that it will receive 0 million if DOE begins accepting waste by 2010 as currently scheduled, and up to 0 million if waste acceptance does not begin until 2015. Funding for the nuclear waste program comes from two sources. Under the FY2005 budget request, 9.0 million would be appropriated from the Nuclear Waste Fund, which consists of fees paid by nuclear utilities, and 1.0 million from the defense nuclear waste disposal account, which pays for disposing of high-level waste from the nuclear weapons program in the planned civilian repository. However, as noted above, the Administration is proposing that the 9 million appropriated from the Nuclear Waste Fund be offset by nuclear waste fee collections. The Administration is also proposing that DOE's Office of Radioactive Waste Management OCRWM ; , which runs the Yucca Mountain Project, take over management of DOE defense and research waste that is currently under another program, bringing the total FY2005 funding request for OCRWM to 7.5 million.
